Titration and settings

• 10 µl AmR - 5 µM final conc. in chamber
• 4µl HRP (500U/ml) - 1 U/ml final conc. in chamber* light intensity of LEDs: 1 mA ( DatLab: Amp polaristion voltage 100 mV) / Gain 1000
• H₂O₂ calibration with a 15.8 µM H₂O₂ solution - 2012-06-20

commercial stock: 880 mM

dilution 1 (1:166.67): 150 µl in 25 ml H₂O = 5.28 mM

dilution 2 (1:333.33): 150 µl in 50 ml H₂O + 500 µl 1 mM HCl = 15.8 µM

→ calibration: 2 * 5 µl in 2 ml chamber = 2* 0.0395 µM = total 0.079 µM (= 79 nM)

• new H₂O₂ calibration with a 28.16 µM H₂O₂ solution, valid from from 2012-06-22:

commercial stock: 880 mM

dilution 1 (1:125): 200 µl in 25 ml H₂O = 7.04 mM

dilution 2 (1:250): 200 µl in 50 ml H₂O + 500 µl 1 mM HCl = 28.16 µM

→ calibration: 2 * 5 µl in 2 ml chamber = 2* 0.0704 µM = total 0.1408 µM (= 140.8 nM)
