MiPNet discussion forum: cytochrome c source (2021-07-16)

Question (US CA San Diego Patel HH)

Cytochrome c (from equine heart) from Sigma Aldrich used in high-resolution respirometry to test the integrity of the mitochondrial outer membrane is currently out of stock for a few months. Does anyone have experience using cytochrome c from another source (e.g. from bovine heart)?

Carolina Doerrier, AT Innsbruck Oroboros

For high-resolution respirometry, we only have experience with the cytochrome c listed on the Bioblast page. For different reasons, we think that it should not be any problem to use cytochrome c from bovine heart. First, cytochrome c is a highly conserved protein across different species, and second, Sigma Aldrich provides the same specifications for C7752 (cytochrome c from equine heart) and C2037 (cytochrome c from bovine heart).

Francoise Van den Bergh, US MI Ann Arbor Beard DA

Our lab uses cytochrome c from bovine heart (Sigma Aldrich: C2037). For the preparation of 4 mM cytochrome c, we follow the information from MitoPedia. We buy the 50 mg bottle, so we do not need to weight it, we just add 1 mL H2O to the whole bottle, and then we aliquot it.
