Manuscript accepted for publication in PloS One, March 9

  • "Physical fitness and mitochondrial respiratory capacity in horse skeletal muscle" (by Votion DM, Gnaiger E, Lemieux H, Mouithys-Mickalad A, Serteyn D) was accepted for publication in PloS One.
  • From the anonymous referee's comments:
    • Reviewer #1: "This is an original work performed with state of the art methodologies, as high-resolution respirometry. The results indicate that not only mitochondrial capacity is a limiting factor of horse performance but also the modalities of energy substrate utilization. Horse is an animal of choice for energetic studies since it shows a huge consumption of oxygen during muscle contraction, suggesting an positive adaptation of the mitochondrial system as compared to other animals. .. This article provides new findings on the link between mitochondrial capacity and whole animal body size and aerobic capacity. It also clarifies whether mitochondrial capacity is a controling factor for horse performance. An important finding is also that the control by the phosphorylation system is stable and does not vary with the performance of the horse. In contrast, the relative utilization of complex I versus complex II varies witht he performance, and this is a new finding which adds up to our knowledge of energy regulation. This work is of broad interest for sport physiology, metabolic researchers and aging research."
    • Reviewer #2: "The results could be described in a much easier way if utilizing classical definitions of mitochondrial respiratory parameters (from the late Britton Chance's milestones), such as state IV (or II), state III, uncoupled respiration, respiratory control ratio and so on." (Comment: Glossary: Respiratory states)

  • "Mitochondrial respiratory control and early defects of oxidative phosphorylation in the failing human heart" (by Lemieux H, Semsroth S, Antretter H, Höfer D, Gnaiger E) was accepted for publication in Intl. J. Biochem. Cell Biol.
  • From the anonymous referee's comments:
    • Reviewer #1: "The manuscript by Lemieux and colleagues offers interesting insight into the bioenergetics of early phases of heart failure."
    • Reviewer #2: "This is a beautifully written work, replete with well-designed experiments and, within the space limitations, a nice review of the pertinent literature. The demonstration of impaired mitochondrial fatty acid oxidation in early stages of human failure is very important, as is the demonstration of defects in the phosphorylation system and Complex I-linked respiration."
    • Reviewer #3: "The manuscript belongs literally to the symphony of bioenergetics and have mastered on a maximum thorough and detailed level a priority study of human heart disease bioenergetics in an intermediate state of heart disease, which was done only scarcely in medical sciences and for this orthodox approach this work deserves a high attention. It elegantly describes detailed oxidative phosphorylation parameters in heart disease compared to the control human heart. Using high-resolution respirometry of permeabilized human heart muscle fibres authors progressively describe distinct aspect of measured data in logical steps to present important differences of diminished mitochondrial function during heart failure. Using elegant protocols developed for years, authors distinguish between phosphorylating (OXPHOS) and non-phosphorylating (ETS) conditions with both complex I and II input using physiological substrates of glucose and lipid oxidation. With presented protocols, authors are able to provide conclusions regarding, e.g., complex I impairment, phosphorylation system limitation (3.2) mitochondrial content (3.3), leak respiration involvement, etc., besides OXPHOS and ETS capacities. Authors use their own terminology describing respiratory states. It would be useful to explain briefly their terminology also in terms of the canonical state 2 - 4 terminology. In spite of this, the methodology approach and presented data are well explained."
