Preparation

Preparation of 10 mM Dithionite solution for Instrumental Background (dissolved in Phosphate buffer, 50 mM, pH 8)
  1. Weigh 0.017 g Dithionite, sodium hydrosulfite, and transfer to 10 mL volumetric glass flask.
  2. Adjust final volume to 10 ml with Phosphate buffer and keep closed to avoid decomposing.
Note: Sodium dithionite cannot be stored for a long period of time, because it is slowly oxidized by air when in solution. Even with the absence of air, solutions of sodium dithionite deteriorate. Therefore when Dithionite is open for a long time, we use a concentration of 30 mM for the Instrumental Background. The experiments showed the identical results with 10 and 30 mM Dithionite stock.
