Description

Selective permeabilization of plasma membrane or isolation of mitochondria are a requisite for OXPHOS analysis since several mitochondrial substrates (e.g., ADP, cytochrome c) are not permeable to the plasma membrane. Plasma membrane permeabilization with effective wash-out of free cytosolic molecules including adenylates, substrates, and cytosolic enzymes makes externally added compounds accessible to the mitochondria. Plasma membrane can be permeabilized mechanically (e.g., liver tissue) and chemically by mild detergents (i.e., digitonin or saponin).
