A principle goal of MitoEAGLE working group #2 is to work towards a human skeletal muscle data repository such that the contribution of Evolution, Aging, Gender, Lifestyle, and Environment to mitochondrial function can be empirically evaluated. A first step toward this goal will be a rigorous and comprehensive literature review of published findings on human skeletal muscle mitochondrial function. Herein, we propose a draft outline for the manuscript to facilitate critical discussion between WG2 members, with the goal of refining the draft and setting milestones for its development to a final manuscript. The manuscript can be broken down into sections focusing on muscle mitochondria and the influence of 1. Exercise/PA/Physical Inactivity, 2. Aging, 3. Gender, and 4. Nutrition and body composition. Particular focus will be placed on differences between methodologies (sample prep, assay protocols, buffer recipes) and how technical variation between studies might confound the biological signal. It is our goal to produce a state-of-the-art review that will inform feasibility of constructing a human skeletal muscle data repository and will also be useful for any investigative teams planning to conduct large multi-site investigations of muscle mitochondrial energetics.
