Description OROBOROS Oxygraph-2k (O2k) - the modular system for high-resolution respirometry (HRR). The O2k is the 2-chamber high-resolution respirometer for monitoring oxygen consumption with small amounts of biological samples. The modular O2k-concept yields a high flexibility for HRR, with the O2k-Core as the basis, and O2k-Modules supported by the O2k-Core. O2k-Modules are the Titration-Injection microPump (TIP2k-Module) and O2k-MultiSensor Modules (O2k-Fluorometer; Ion Selective Electrodes, ISE)

O2k-Core OROBOROS O2k-Core - the experimental system complete for basic high-resolution respirometry (HRR). The O2k-Core includes the O2k-Main Unit with stainless steel housing, O2k-Assembly Kit, two OroboPOS (polarographic oxygen sensors) and OroboPOS-Service Kit, DatLab software, the ISS-Integrated Suction System and the O2k-Titration Set. The O2k-Core supports all add-on O2k-Modules of the O2k, and is a component of the O2k-Fluorometer.

On-line display of oxygen flux (rate of respiration) is provided in addition to the conventional 'oxygraphic' plot of oxygen concentration over time. Highest signal stability minimizes the required amounts of biological sample, and provides the basis for resolution in the extreme low-oxygen range. Peltier temperature control provides a thermal stability at ±0.002 °C in the range of 4 °C to 47 °C at typical constant room temperature. Electronically controlled PVDF or PEEK stirrers are integrated in the two-chamber design of the O2k, and a barometric pressure transducer enables automatic oxygen calibrations implemented in the DatLab software.

The O2k is a sole source apparatus with no other instruments meeting its specifications.

Power-O2k Core-Unit: The Power-O2k Core Unit is an economical option for using additional O2k-Units to increase output in high-resolution respirometry. This application does not require an additional ISS-Integrated Suction System, an O2k-Titration Set and the O2k-Manual. Furthermore, the following OroboPOS Service Tools can be used from the available O2k-Core and are not included: OroboPOS-Mounting Tool, OroboPOS-Polishing Cloth, OroboPOS-Polishing Powder and Pen-Contact Oil.

The Power-O2k Core-Unit supports all add-on O2k-Modules of the Oxygraph-2k.

O2k-Fluo LED2-Module The O2k-Fluo LED2-Module is a component of the O2k-Fluorometer. It is an amperometric add-on module to the O2k-Core (Series D upwards), adding a new dimension to HRR. Optical sensors are inserted through the front window of the O2k-glass chambers, for measurement of hydrogen peroxide production (Amplex red), ATP production (Magnesium green), mt-membrane potential (Safranin, TMRM), Ca2+ (Calcium green), and numerous other applications open for O2k-user innovation. The O2k-Fluo LED2-Module consists of optical sensors for both O2k-Chambers (LEDs for green and blue excitation), optical filters, Fluorescence-Control Unit for regulation of light intensity, data input into the O2k-Main Unit, and the updated DatLab software. 12100-01

O2k-TPP+ ISE-Module O2k-TPP+ ISE-Module: 2 potentiometric ion-selective electrodes (ISE), ISE-Service Kit and 2 PEEK-Stoppers, supported by the O2k-Core. The ISE module is designed with replaceable membranes making it possible to measure different ions such as TPP+ or Ca2+ with the same electrode housing. The main application of the O2k-TPP+ ISE-Module is measurement of mitochondrial membrane potential with tetraphenylphosphonium (TPP+) or TPMP+. 12300-02

ISS-Integrated Suction System ISS-Integrated Suction System: Suction pump 230 or 120 V with stainless steel housing, 2 liter waste bottle, filter and tubing; for siphoning off excess medium from the O2k-Stopper and for emptying the O2k-Chambers. The ISS is included as a standard component of the O2k-Core. Media containing living cells or microorganisms, various poisons (inhibitors, uncouplers) and mixtures of proteins and substrates are safely disposed off in the 2-litre waste bottle.

The suction pump (230 V or 120 V) is enclosed in a stainless steel housing, which holds a readily accessible mains switch, two removable receptacles for the tip of the tubing, an easily removable and safe connection to the gas filter which further connects to the fully stabilized waste bottle. The waste bottle with filter can be simply removed for emptying, cleaning and drying.

O2k-Titration Set The O2k-Titration Set consists of Hamilton microsyringes (6 x 10 mm3 and 3 spare plungers, 6 x 25 mm3, 2 x 50 mm3, 1 x 500 mm3), provided in the Syringe Storage Box with Syringe Labels, a set of two Syringe Racks with Syringe Collars, and a set of two Tube Racks. OROBOROS O2k: development and distribution

The O2k is developed by OROBOROS INSTRUMENTS in cooperation with WGT, produced in Austria by WGT, and distributed world-wide by OROBOROS INSTRUMENTS.

The O2k-Modules can be combined in any configuration with the O2k-Core. The modules can be added any time to extend the Oxygraph-2k as an O2k-MultiSensor System according to application-specific objectives.

Auxiliary HRR-Tools provide extended support in the context of high-resolution respirometry: PBI-Shredder HRR-Set for tissue preparation; Microbalance, HRR-Dissection Set.
